標準液配制方法：

取400NTU的標準原液50mL注入容量瓶內(nèi)，再取950mL純水注入容量瓶內(nèi)、搖晃混勻，此液體為20NTU標準液。

二、10NTU 標準液配制方法：

取400NTU的標準原液25mL注入容量瓶內(nèi)，再取975mL純水注入容量瓶內(nèi)、搖晃混勻，此液體為10NTU標準液。

三、配制的10NTU及20NTU標準液，要現(xiàn)配現(xiàn)用，常溫下存放，就會變化，不能使用。

四、400NTU 標準液，不能在常溫下存放，濁度標準液應(yīng)貯存于1—10℃以內(nèi)環(huán)境中(放置冰箱冷藏室內(nèi)貯存)。

標準溶液配制：

1。先將PH7.00同PH4.00藥粉分別倒入250毫升蒸餾水中，充分溶解，用兩支玻璃瓶密封保存，做好標簽。

2 。在校正前，準備ORP標準溶液。

3 。86毫伏：取50至100毫升PH7.00標準溶液置入一玻璃杯中，再加入稍許醌氫醌，充分攪拌。

4 。256毫伏：取50至100毫升PH4.00標準溶液置入一玻璃杯中，再加入稍許醌氫醌，充分攪拌。
5。 ORP標準液保存期為72hour。

注意事項：

1.流動相必須用HPLC級的試劑，使用前過濾除去其中的顆粒性雜質(zhì)和其他物質(zhì)(使用0.45um或更細的膜過濾)。

2.流動相過濾后要用超聲波脫氣，脫氣后應(yīng)該恢復(fù)到室溫后使用。

3.不能用純乙腈作為流動相，這樣會使單向閥粘住而導(dǎo)致泵不進液。

4.使用緩沖溶液時，做完樣品后應(yīng)立即用去離子水沖洗管路及柱子一小時，然后用甲醇(或甲醇水溶液)沖洗40分鐘以上，以充分洗去離子。對于柱塞桿外部，做完樣品后也必須用去離子水沖洗20ml以上。

5.長時間不用儀器，應(yīng)該將柱子取下用堵頭封好保存，注意不能用純水保存柱子，而應(yīng)該用有機相(如甲醇等)，因為純水易長霉。

6.每次做完樣品后應(yīng)該用溶解樣品的溶劑清洗進樣器。

7.C18柱不能進蛋白樣品，血樣、生物樣品。

8.堵塞導(dǎo)致壓力太大，按預(yù)柱→混合器中的過濾器→管路過濾器→單向閥檢查并清洗，清洗方法；①以異丙醇作溶劑沖洗：②放在異丙醇中間用超聲波清洗；

9.氣泡會致使壓力不穩(wěn)，重現(xiàn)性差，所以在使用過程中要盡量避免產(chǎn)生氣泡。

10.如果進液管內(nèi)不進液體時，要使用注射器吸液：通常在輸液前要進行流動相的清。

11.要注意柱子的PH值范圍，不得注射強酸強堿的樣品，特別是堿性樣品。

12.更換流動相時應(yīng)該先將吸濾頭部分放入燒杯中邊振動邊靖洗，然后插入新的流動相中。

對照品實驗方法與判定：

1.對照品溶液的穩(wěn)定性

2.對照品溶液的配制：精密量取腦對照品適量，加無水乙醇制成每1ml含腦1mg的溶液，作為對照品溶液。

3.色譜條件：以交聯(lián)鍵合聚乙二醇為固定相的毛細管柱；柱溫120℃；進樣口溫度250℃；檢測器溫度250℃；分流進樣，分流比10:1。理論塔板數(shù)按腦計算應(yīng)不低于10000。

4.實驗方法：配制的對照品溶液，一份置冷處保存，一份置室溫中保存，在第1、3、7、10、15天重新配制一份對照品溶液，三種儲備液同時稀釋制成每1ml中約含50ug的溶液。照氣相色譜法，并依據(jù)新對照品的峰面積計算原對照品溶液的含量。記錄下來，保證原對照品溶液含量在考察期相對平均偏差不超過2.0%，驗證對照品溶液在使用期內(nèi)穩(wěn)定可靠。
